As far as setting up your mac to maximize it's efficiency, it comes down to personal preference. Out of the box, I left my mac the way it is. About the only tweak I made was providing materials that were not given to me, such as a surround sound adapter and such. I use the mouse on ocasion, when VoiceOver will not do the prefered mouse action I am looking for. Overall though, I left Safari the way it was, and only enabled the universal access help. To find this, go to Safari, and while in Safari, press and hold down the command key, and then press the comma key. This will bring up your preferences. Do vO + Home, to Navigate to the top, should you not be there already. VO + Right Arrow until you hear Toolbar. Interact with VO + shift + Down, and then VO + Right arrow, until you hear Advanced. VO + Space, and uninteract by pressing VO + Shift + Up. Afterwards, you will use VO navigational keys, and the first area you will come accross is Universal Access. Navigate until you hear, Press Tab to highlight each item on a webpage. VO + Space on the check box, and this will be enabled. This will allow you to tab through links, and such on a page, just like other screen readers you may be familiar too.

Other then that, search engines, and taking things off Safari is always up to you. My personal preference is to leave everything as is. I like the way it was already presented to me, so I have no desire to change it.

Yes VoiceOver has a way to go to be as advanced as Jaws for example but Apple really haven't done bad. Anyway as for what you asked, I can not really comment on mouse as I am totally blind therefore don't use the mouse!!! As for Safari I have it set to Google however I'm sure you could set it up to start with other search engines, I think this is down to personal preference though.
