With the Airport software running it should offer the choice of
"Connect via: Airport PC Card". I doubt it matters which card as
long as it's one that works with WaveLAN / Orinoco / Lucent / Airport
drivers. When you tried the Airport software did you start in the
Airport Tool under the Apple Menu. Even with the Airport card turned
off it still lists it as an option under "Connect via:"
No, turning the USB stuff off shouldn't affect things.
