The situation is confusing, indeed.
Here my latest experience.

With the 2.6.19 kernel but the wacom driver from Linuxwacom 0.7.7.6  the
'testinput' routine from a gtk+-2.10.9 does work fine with my
ps/2 mouse and my Wacom Graphire 4 (USB) (with pressure sensitivity).
It seems as if very recent versions of Gimp (SVN last weekend)
have ameliorated the situation a bit.

It looks like an initialization problem anywhere in either Linuxwacom
or GTK+.

The following has been reproducable so far.

- Gimp and Inkscape both have problems if the extended input
  devices are enabled from the very beginning.

- With most recent versions of Gimp, just using the
  Wacom pen a single time in the canvas, enables both,
  the mouse and the pen. (Earlier, I had to disable the
  extended input devices, use mouse and pen (this time
  without pressure sensitivity) in the canvas,
  then reenable the   extended input devices and
  suddenly both work fine (with pressure sensitivity)

- If (after startx) testinput has been invoked once,
  both, Gimp and Inkscape work just fine with pen and mouse
  from the outset.

It smells as if there is something uninitialized in the
Linuxwacom driver(s) or GTK+, which gets initialized if the first
application is 'testinput' from the GTK+ package.

So, hopefully we see some convergence soon,
