I sometimes have an identical problem on my brand new iBook, so I suspect it might be software-related with iTunes. I select the song to play, the pointer moves (indicating the song is playing), but there is no sound. I can sometimes get sound again by playing a sound in another app like Quicktime, then switching back to iTunes. Otherwise, I have to restart iTunes, and one one or two occasions, I've had to log out and back in to OS X.
