Yes, you can do so. If you are in the middle of a shooting and find out you
can't stop to clean the thing, then you better make this back up shot. If
you have the sample of clean blue tropical sky I will be sending Paul, or
just another clean ,evenly colored  area you can shoot, you can then assume
the pattern of dust remains the same throughout the entire shooting. THen
you go home with your files and add the sky shot as a second layer and  use
blending modes to eliminate the uniformly colored background, and all will
be left  in the layer is the "template" of dust spots to be used for further
retouching.

That is exactly why I said battery-operated Vacuum thing.

If you use an industrial sized machine and If the chip is sucked from your
camera, Canon/Nikon  will not accept any liability nor will I !!

 Others have  reported good results with Hair Blowers too. Perhaps a mix of
both blowing and vacuuming will do wonders to hard-to-remove dust spots.

I was a chemist in my former, pre-photography life and I keep very litle
respect to Canned Air, Canned CO2 or  liquid chemicals for cleaning such
sensitive surfaces,regardless of their claim for chemical inertia.
 Remember that the dust is not falling on the chip, but on the IR/Low Pass
Filter in front of the chip.  A scratch  or a clouding of any size in the
filter screen means sending your camera to repair at full price, with all
warranties void.
