It can, yes.  You want to leave a mouse configured in /etc/X11/xorg.conf, but 
with the following so X won't complain about there not being a mouse plugged 
in when it starts.

Section "ServerFlags"
        Option          "AllowMouseOpenFail"    "true"
EndSection


It won't cause any issues with day-to-day usage (and in fact if you plug a usb 
mouse in for whatever reason, it should just start working immediately), but 
the wacom driver seems to lose most of its features if your tablet is the 
only configured input device.
