The command comes from SqueezeCenter,  in fact ALL commands come from
SqueezeCenter.  iPeng just communicates with SqueezeCenter.  Seems like
all you need to do is have a script in SqueezeCenter that sends your
command whenever a specific player is turned on, cause you really do
not need to turn on the speakers independent of the player attached to
it right?

Another option is if the speakers can be triggered via IR, you could
use the IRBlaster plugin and connect an IR transmitter to your SB3
headphone jack.
