You don't select the edb file by itself. If you are currently backing up the mailboxes (referred to as Brick Level), then you certainly should have the ability to back up the entire database. What you will need to do is in the same section that you select the individual mailboxes, select the Information Store (or Storage Group if you are Exchange 200x).
