Andy,
To restate my previous comment,
When using unsubscribe via subject line users must be "exact".
If passwords are involved then it must be in the form
[:password:]unsubscribe
or [:password:]remove, etc. without a space between :] and unsubscribe
also with no trailing text following unsubscribe.

In other cases, with out passwords and with or without poster.lst
users should only type: unsubscribe
with no leading spaces or trailing text like: unsubscribe [EMAIL PROTECTED]
So, if your list is not using passwords just have them
type:  unsubscribe
on the subject line only and see if that works.
