Silver-through-hole (STH) is a fabrication process - not a PCB layout process. The fab applies a silver conductive paste through the via holes instead of copper plating the holes.

Discuss with your fab what they can do for you with regard to via hole sizes using STH. Then lay out your board accordingly. Nothing special is required in the PCB file - except, perhaps, a note on your drill drawing specifying that you want STH.
