No - The trial mode is not one of the options on the toolbar. As stated before, there was a second toolbar in 4.5 that was automatically generated and had the trial mode. The additional toolbar in "options" was also in 4.5 but that is a different matter. The trial mode is important because it is useful to run variations to explore ideas without actually including them as a pgn variation. Also, I note that when entering a variation, I am not asked if I want to replace the main move (even when I have selected "ask me" as an option). Rather, I need to go a menu below the board and change it to the main line there. Still, I like the interface and improvements in 4.6. It just needs some functionality tweaking before I can replace 4.5.
