One suggestion that I haven't seen being made yet.

Be certain that no one installed any software, or updates, on the machine 
that may have replaced the database drivers.  When this happens it tends to 
cause problems.  Refer to the Protel KB for details on how to repair this 
by re-installing the proper drivers.

Since there appears to be an issue with creating new DDBs I suspect that 
this is a possibility.

It's also possible that some software update/installation also replaced a 
shared DLL, but I'd shoot for the database drivers first as that's an easy 
one to try.
