Hi John,
I would suggest following to troubleshoot:
TRY SERIAL CONNECTION
1) Check your serial cable. The recommended cable in the User Guide is
"Apple System Peripheral-8 cable (Part Number M0197)".

1A) As a really rough check on whether you have a connection, boot up
in OS-8 or 9, with the printer connected via the serial port, the
printer powered on, Appletalk on, and select "connect via printer
port" in the AppleTalk control panel. The printer should start
printing random characters. If it doesn't, one might suspect either
the cable itself, or the interface hardware at one or both ends of
the cable.
1B) If possible substite a cable which is known to be working from
another installation. If this is not possible, you can manually
check each pair of pins in the cable using an ohmmeter and the wiring
diagram from the above link. Inspect the cable visually for broken,
bent, missing, or 'pushed in' pins.
1C) Assuming the cable is OK, try to eliminate corroded or dirty
connector pins and/or sockets as a possible cause. Visually inspect,
seat and re-seat the cables.
2) if you are happy that the cable is good, and given that your USB
to Serial adaptor is also not working, suspicion can fall on either a
hardware problem at the printer end, or a software problem on the
Mac. I think the easiest check of the software would be to download
the latest SC600 software from Epson, then restart with the base set
of extensions and control panels, using the Extensions Manager
control panel, before reinstalling the drivers. Do this on both Macs.
3) If 1) and 2) have failed to produce a result, you'd have to start
taking the printer apart and inspecting the circuit boards for
possible problems, or else suspect that both your Macs have faulty
serial ports.
